      cathepsin C在LPS 誘導的小鼠腦內神經炎癥中表達的變化

      摘要:組織蛋白酶C(cathepsin C)是免疫細胞中絲氨酸蛋白酶的激活劑,參與人體多種病理生理過程,但是cathepsin C 在中樞神經系統神經炎癥中的改變至今未見報道。本研究采用C57BL/6J 小鼠腹腔注射LPS (5 mg/kg),建立神經炎癥動物模型。利用免疫組化和原位雜交方法檢測注射后腦內小膠質細胞的活化,炎性因子以及cathepsin C 的表達。結果發現,LPS 注射后腦內小膠質細胞活化,炎性因子表達明顯,cathepsin C 的表達顯著增強,見于多個腦區的小膠質細胞中。上述結果提示,在LPS 誘導的神經炎癥中cathepsin C 的表達顯著上調,cathepsin C 可能介導了神經炎癥反應的發生發展過程,為進一步研究其在神經炎癥
      中的意義提供了形態學資料。

      The changes of cathepsin C expression in LPS-induced neuroinflammation in mouse brain

      Abstract: Cathepsin C functions as a central coordinator for activation of many serine proteases in inflammatory cells. cathepsin C is responsible for many inflammatory diseases. In this study,C57BL/6J mice were injected with lipopolysaccharide (LPS, 5 mg/kg i.p.) or saline. Immunohistochemistry (IHC) and In situ hybridization(ISH) methods were used to examine the activation of microglia, the expression of inflammatory cytokines as well as the expression pattern
      and cellular localization of cathepsin C in the brain. The results showed that after LPS injection,microglia was activitated, inflammatory cytokines were expressed significantly, and cathepsin Cwas expressed in microglia in a global pattern. The results indicate that cathepsin C could participate in neuroinflammation induced by LPS.
